I don't believe that Jesus Himself specifically spelled out the answer to your question in the passage cited. However, as I see it, Jesus was speaking to people who did not recognize or accept Him as the Messiah sent by God. After Jesus ascended to heaven (forty days following His resurrection), such people would seek Him (in the same way that they came to hear Him in the temple) and not find Him, because, in this life, He would no longer be visibly present on earth, and because those people would also not be with Him in God's kingdom in eternity, due to their lack of faith in Him as the means provided by God to gain forgiveness of sins and eternal life in God's presence.
