The Lord had spared the first born of Israel when the Egyptian first born children were destroyed when Pharaoh refused to let the children go. Appropriately the firstborn of men and animals were to be "set apart" for His service.
Since God killed the firstborn of the Egyptians, and spared the firstborn of the Israelites, He ask them to sanctify all firstborn as a gift for Him not killing their firstborn. It also signifies God's Firstborn Son who was sanctified and set apart for God's use and purpose.
