As I read the cited verses, the "mystery" to which Paul is referring pertains to something that was previously hidden, but has now been revealed. It is a twofold revelation, referring not only to the ability of Christ (now that He has been incarnated and also completed His atoning work and returned to the Father) to both sanctify and dwell within believers; but also to the expansion of the body of those believers to include not only Jews, but also Gentiles (such as those to whom Paul was writing),

Based on 2 theological dictionaries, Eastons's Bible Dictionary and International Standard Bible Encyclopedia, the "mystery" in Colossians 1:27 and Col 2:2-3 is not a riddle or something impossible to understand. It is a gospel truth, once hidden in God's plan, but now publicly announced: The mystery is revealed (Col. 1:27) which is this: Christ Himself living in you, the Gentile believer, which is your absolute guarantee of future glory with God. The content of the mystery (Col. 2:2-3) is this; Christ Himself is the ultimate treasure chest containing all the wisdom and knowledge you will ever need. You do not look anywhere else. So, the "mystery" is not a what but a who: Jesus Christ, revealed as your indwelling Savior and your complete source of wisdom.
